Date: 30 December 2008; Time: 1400; Agenda: Quantification of 72hr biofilm at 1 temperature and biofilm formation for 24hr and 72hr, 30deg.

Quantification of biofilm:

• This time we quantified the biofilm formed based on only CV.

Biofilm formation:

• Similar procedures carried out to grow biofilm, only that now they are left to grow for 24hr and 72hrhr at 30deg. 30rpm. Also, we now use a new COLOURLESS M9 as previously prepared M9 may have been prepared wrongly as we autoclaved it in the last step after adding glucose --> may have resulted in decomposition (hence the color change). For this new colorless M9, we did not autoclave in the last step.
• Columns 1-6: M9 culture(centrifuged); Columns 7-12: LB culture
• Similar plating done to verify McFarland standards.
