## Advance Preparation

Recall that by conscientiously working in the lab, you have been preparing for the lab certifications all along. Nevertheless if you want to "get ready" for the lab certifications, review your lab notes and notebooks. Use the wiki to clarify what seems cryptic. You will have your notes, notebooks and calculators but not your computers during the lab certifications.

## On the day of the first lab certification

- This certification will take place during the first hour of lab on M1D1, 9.11.12 or 9.12.12.
- You will have the print-out of the lab tour to work with but not the computers.
- You will have to show your work for any calculations you perform, though you can use a calculator for computation.
- You and your lab partner can work together on the questions but you should turn in answers individually.
- This lab certification is worth 2% of your final grade.

## On the day of the second lab certification

- This certification will take place during lab on M1D6, 10.2.12 or 10.3.12.
- You will have your laboratory notebook -- update: and pre-lab notes -- to work with but not other notes or computers.
- You will have to show your work for any calculations you perform, though you can use a calculator for computation.
- You will work alone on this lab certification, not with your lab partner.
- All the Mod1 content from Day 1 through Day 6 is fair game.
- The lab certification is expected to take approximately 1.5 hours.
- If you've been keeping up with your lab notebooks, then you should have easy access to entries:
- describing the PCR you did, including the basic design elements for primers
- with information about the transformation you performed
- with data and the logic behind your diagnostic digest
- with some calculations and data analysis associated with ligations, transformations
- with protocols for plasmid minipreps
- describing restriction analysis through agarose gel electrophoresis.

- This lab certification is worth 8% of your final grade.
